About the Aplication Prefs / General: Coordinate Settings Never noticed but the QCAD 3.20.1 UI does not format the number as set in the prefs. I cannot reproduce the problem with the semicolon. QLCDNumber has a property called digitCount:
http://doc.qt.io/qt-5/qlcdnumber.html#digitCount-prop

So if w if your LCD widget, you can simply do:
w.digitCounter = x;

Die Länge der Linie kann oben in der Werkzeugleiste eingegeben werden:

Wird statt Liniensegmenten eine unendliche Linie oder ein Strahl erstellt, so ist die Länge irrelevant, da diese Linien unendlich lang sind.

Misc > Import/Export > Import Points imports points from a simple x,y list:

x1,y1
x2,y2
x3,y3
...

However, it might be more convenient to simply paste your x,y list into the QCAD command line input:

Layer "Defpoints" is an internal layer of the DXF/DWG format. It is by definition not plottable. Long story short: don't use layer "Defpoints" for anything and just leave it as it is.
